For me it's gotta be "Bill the Butcher" played by Daniel Day-Lewis in Gangs of New York.
It's not that it's my favorite movie, but i do like it a lot. But that was an amazing performance. I just can't think i've seen any better.

Robert Downey Jr makes a great Tony Stark. I don't know if it's my favorite, but it's the first thing that came to mind.
Maybe Harrison Ford as Indiana Jones - he made the role into a classic character that I don't think would have worked nearly as well with any other actor.
And maybe Johnny Depp as CAPTAIN Jack Sparrow. In the first movie in particular, instant classic.

---"This week, it’s Jack Lemmon in “Glengarry Glen Ross.”" Pure subtle brilliance in that one.
I gotta go with Peter Sellers in Dr. Strangelove. Not subtle at all!

I think my favorite role/performance executed with perfect casting is Ian McShane as Al Swearengen on the HBO series Deadwood. Mesmerizing.
