(1) Put on an aftermarket Telly imitating bridge PuP like a Stelly.
(2) Put in pull pot wired to give you bridge and neck.
(3) Notice movement in the right tonal direction, but bemoan the fact that you can still tell it's not a Tele.
(4) Buy real Tele.

Rewire one tone control as a blender pot to add either the bridge or neck pickup to whatever is currently selected. You can get neck and bridge pickups together, with the added bonus of all three pickups together too.
It doesn't sound exactly like a tele but it's a great sound.

Use bridge pickup.
Use middle pickup.

Use the bridge, or middle pickup, tone on 10, 9, or 8. Play the strings closer to the bridge pickup.
Similar tone but not exactly a Tele. But it is in there. Experiment till you find it.

Start with a hard tail Strat. The vibrato bridge softens the attack and spreads the vibrations out across the vibrato springs and all the strings.

I have tried this below, with a noiseless bridge pickup (Mid & neck are 57/62 reissue pickups) . I have ordered a copper-plated base from stewmac, they sell DYI pickups kits. I have used wax to stick the brass place to the noiseless pickup, with some glue on the edges.
After 2 years with this setup I bought a hot rod telecaster with a Seymour Duncan Jerry Donahue at the bridge. My Strat with this setup sounds close to my Tele at the bridge, but there is nothing like the real thing !

I asked to myself the same question for a couple of years, trying to make my strat to sound as (near) a tele. Stoped trying as soon as I got a Squier CV Custom 60 Telecaster.
It's cheap, plays and sounds very well and it souds like a... Tele. No more time waisted trying to make my strat sounds like a Tele because just a Tele sounds like a Tele!
Do yourself a favour: get a Telecaster.

You can also buy a Deluxe Player's Strat for around 800.00. This guitar has the wiring setup to engage neck and bridge pickups for that middle position Tele like tone. Very pretty sound with great midrange in that position. Also give all 3 pickups on for a warmer out of phase tone.
Deluxe Player's Strats are great guitars for the price. Ash body, gold hardware, vintage noiseless pickups.

Good advice. I once owned one, a 1963. It had a cream color body with a black pickguard. I sold it many moons ago. Pretty stupid selling it but I needed the cash.

A steel, or ferrous plate under the bridge pickup will help. Plating doesn't matter. Fender used a copper plated plate because it was easier to solder to and it kept the steel underneath from rusting.
A note if you have noiseless pickups...if your pickup is a side by side coil pickup, like a rail or GFS Neovin, the plate won't work well.
But on a single coil pickup, the plate widens the area the pickup "sees."
The other factor on a Tele is the material the bridge plate is made of. Some are chrome plated steel (ferrous) and some are plated brass or stainless. A ferrous plate will further spread the pickup pattern. A non ferrous plate (brass, stainless etc) will not. A thin steel plate under the pickguard covering the area of a Tele bridge plate could help towards Tele tone, but I'd do the steel plate under the pickup first.

Thanks Steve. I don't really want to make any mods to the guitar. Just try to get close with adjusting the knobs and switches. I gave up modding guitars long ago (outside of replacing a pickup once in a blue moon).

Wire a mini toggle switch for less than two bucks. With the switch engaged the neck pup is always on; giving that elusive neck-bridge Tele tone and some other interesting combinations. Both of my partscasters are equipped this way. Blocking the trem makes sense too.

Wiring the bridge and neck in series, rather than the standard parallel will get you close. Also wiring the bridge and middle in series and in parallel with the neck will get you close as well.YMMV

All the wiring mods above seem good alternatives for the bridge and neck tele pups together but what about the bridge tone alone?
To me, the typical telacaster tone is not only recognisable when using both its bridge and neck pickups together, but the the bridge itself, alone, is a vey distinctive telecaster tone.
For that reason, again, just a telecaster will give you the real characteristics Telecaster tones and as you know, you can get a Squier Classic vibe new for not much money and a used one for very few $$$. Same thing for a used MIM Tele.

The Tele has the more unique bridge tone. That is correct. However, that is providing that you have a good bridge pup with a base plate and a good bridge to transfer tone.
I like the vintage ashtray bridge for that reason. The pickup with it's metal base plate sitting in that bridge assembly is what really defines the tele bridge tone. I also like 3 brass saddles but are those who prefer to have 6 for more accurate intonation adjustment.

A Tele and a Strat will never sound alike.
You can dial the Strat in close (on a tube amp) by increasing treble on the amp and using a compressor pedal as a preamp.
